Hello. I will be extracting RNA from frozen eucalypt pollen soon. It has been in the -80 freezer for over a year. Should I rehydrate it before I extract RNA? I will also be germinating some in-vitro. Should I rehydrate the pollen before I germinate it?
I welcome and appreciate any advice in the matter.

We have done pollen germination on solid germination media and it works well, but I plan on doing germination in liquid media. The few times I have tried to do germination in liquid media, it has not worked as well as on solid media. This is why I was wondering if I should hydrate the pollen prior to pollen germination in liquid media. We plan on using liquid media instead of solid media because we can centrifuge the germinated pollen grains and extract RNA right away. Using solid media will complicate and delay the RNA extraction.

Thank you so much for your advise. Do you think I can just shake the tubes slowly to aviod damaging pollen tubes?
