I want to make a recombinant m13 phage displaying my peptide as a fusion peptide on its surface pVIII. Should i add ATG at the beginning when i want to insert it in phagmid vector? Previously, a protein which already had ATG codon was inserted and displayed using this system. As my 101 peptide lack ATG the +2 ORF does not exist anymore when i check the vector in snapgene.

If I understand your experiment correctly, your plan is to add (append) your peptide to pVIII or insert it. If your vector has everything to express pVIII, then you don't need an extra start codon. Could you re-post the vector map in a higher resolution, please?

The peptide which was previously inserted in this phagemeid was SIN-Beta2 microglobulin. It contained an ATG codon at the beginning of sequence and was translated in +2 ORF. In attachment you can find the complete map and sequence of this vector containing beta2microglubin insert.
