Can anybody please suggest which analytical technique would give me the best result for the size and shape of solid lipid nanoparticles SEM or TEM or AFM.
The nanoparticles are synthesized by high pressure homogenization and coated with a biopolymer and spry dried with mannitol.
the powder when redispersed has a size of 5microns.
I need to know the the actual size and morphology of the SLNs.
Please suggest which technique to apply SEM/TEM/AFM.

Since your particles are relatively big, you could use a surface profiler to measure approximately 5 micrometer size particles easily. Then you could use a SEM instrument to double check the results.
According to the dimensions you have given, the particle size is much larger and it is in the micron range. As a result, SEM analysis is suitable for this. From there you can easily measure the actual dimensions.

You can use SEM or AFM techniques to monitor morphology/phase separation and topography of your samples. Honestly, I do not think you need TEM for micro-scale observations.
Both secondary electrons and back-scattered electrons can be used for morphology and phase separation evaluations. However, if your sample is sensitive try no to use high electron voltages. I think about 10 kV would be fine.
Also, you can measure the surface roughness of your samples using AFM. Again, you might want to use non-contact mode to not damage your samples.

All these techniques can be useful. TEM should be the best, however you need to find a good contrast. Be aware that the electron beam can "burn" your sample! In the light of this problem, AFM can be a very useful technique. Let me know which technique you will choose, I'm very curious about which gives the best results in this case!
