For our PCR reactions, we would add wax solution available from Biorad to prevent evaporation and inter-mixing of samples during the run. Since that product has been discontinued, we went around looking for suggestions. I would like to know if anyone tried to use Mineral Oil or anything else? A protocol or reagent prep would be extremely helpful, thank you!!
If your PCR machine has a heated lid (most of the contemporary PCR machines will) then you can exclude this step altogether. If not, then you can definitely replace wax with mineral oil. However, it is more preferable to use paraffin wax, as mineral oil is more difficult in handling (can lead to contamination), and has been shown to decrease the efficiency of post-PCR manipulations. So, yes, you absolutely can use mineral oil, just be mindful that there is more room for error.
