I always get great help from researchgate.
Today I want to ask about cell culture medium.
I work with huvec, studying angiogenesis.
In general, I use m199 for working with huvec but sometimes I use EGM instead of m199. I know that EGM is more efficient for cell growth. I have worked with huvec in 3 cases below.
1) Use of M199
2) Use of EGM and EBM
3) Use of M199 but changes medium(M199 → EGM and EBM) during my work.
Here is my question :
When conducting same specific huvec works(migration assay, sprouting assay, rt-pcr, western blotting and so on) but in different conditions(3cases above),do I have to set steps of protocols differently according to 3 cases? I think it is totally ok for me to follow same steps in all 3 cases above but I want to get confrimed from researchgate.

(For example, I use different medium for starvation process according to three conditions
above)
What I wanted to ask is that whether I have to add or remove some procedures(steps) totally
according to diffrent three conditions in my question when working with huvecs.

However, in the present case, I think that you perfectly address the problem.
An I actually think that you have to perform your standardized experiment with HUVECs growing in the three culture media you describe.
I am ready to bet that you will obtain distinct results with the three different media, but this would be of great scientific interest and it certainly be of added value to your future publication.
I am attaching here a most than interesting article about this topic.
Trabectedin is today a marketed anticancer druh with fantastic characteristics, e.g. one of the first anticancer drug targeting not only cancer cells but also cancer-associated cells!
In 2004, the researchers of the attached article published a major finding about the influence of culture medium conditions on the data you can obtain!!

To make very short a potential very long story, I actually think that you have to perform the experiments you have in mind without REMOVING anything from your protocols ... and the data you will obtain can bring you very good surprises!
I do not think that you have to ADD something, but this is based on my own opinion that must be validated by other RG members.
