The thickness and timing in staining is very crucial in histology.
I use 6 micrometer thick sample and sometimes fast green just need a dip about 3 seconds before it actually over stain the whole sample...
Any better suggestions to replace fast green?
I am looking on general cellular structures with the presence of observable vascular system...
if anyone could provide multiple staining with very less counterstain or overriding then I would appreciate very much!
You might be able to achieve the staining you need simply by diluting the FastGreen further. A lower concentration of fast green would presumably take longer to stain your sections and give you better control over the intensity of your labeling.
David M Sherry , noted with thanks! I will try it out with lower concentration...but what is the concentration you would like to suggest? I used to prepare 0.05 g fastgreen with 100mL 95 % ethanol...then filter it using filter paper or ultrafilter...
I agree with David , you can dilute your prepared stain with 95% ethanol ....one part from your prepared stain to one part 95% ethanol
