I am in need of some help. I am currently exploring the effect of certain peptides on the production of cAMP in rat myocytes. I am using the Perkin and Elmer LANCE cAMP assay and I have managed to produce a nice response to forskolin, but when I try peptides which require GPCR activation I get no cAMP production (even though there should be). I am following the LANCE protocol.
