I was wondering if any one have checked RNA stailbity at different temperature for subsequent PCR amplification. I want to store my RNA isolated from blood vessels before preparing the cDNA.
usually I store the isolated RNA at -20 degree celcius in RNAase-free water if I need to use frequently within 1 month. And I always use DNase treatment before storing the RNA more stably. Although, frequent freezing and thawing may degrade some RNA. But I store the isolated RNA at -70 degree celcius for around 1 year and they are very much stable.
I hope the information will be helpful for you. Good luck with your experiments!
Thanks, Rahul. We performed DNase treatment and eluted the RNA in RNase free water. Hopefully, the eluted RNA remains stable in our freezer as suggested by you.
Dear colleagues, my own experience tell me that even in the case of RNAse free water, in -80oC you can receive reference/target gene shifts. I don't recommend to compare expression between fresh (without storage), -20oC (even 1 day) and -80oC.
