Iam using cisplatin on my HEK293 cells . By using MTT test, cisplatin failed to induce any toxic effect what could be the possible causes? I incubated 100 µM of Cisplatin with my HEK293 for 22hrs.
Hello sorry I know you asked this a while ago! Maybe you have already solved it. What did you use to dilute the cisplatin? DMSO has been shown to reduce the effectiveness of cisplatin. Was the drug within the best before date? Did you make it freshly just before applying- as there are some papers showing cisplatin in solution degrades within one week.
thanks alot for your reply . I actually donot use DMSO as it can reduced cisplatin toxicity. I always use fresh cisplatin solution.
Sorry for the delayed reply. Have you tried leaving the cisplatin on for longer- 72-96 hours? It might take a while to see the biological effect. How did you managed to get a concentration of 100uM? Thats really good! I've been struggling to get cisplatin to dissolve
we take already dissloved cisplatin (1mg/ml) from the pharmacy , which is dissolved in NaCl. which is then diluted by either buffer or culture medium to the desired concentration. I hope that helps
