Dear All,

We are optimizing the in vitro method for "The Catalytic Bleomycin assay of spiked human serum samples". Since most of the available literature referring the article titled with “Measurement of Iron and Copper in Biological Systems: Bleomycin and Copper-Phenanthroline Assays by P. J. EVANS and B. HALLIWELL”. We are trying to reproduce the same method.

We have following questions

1. We are reproducing the method by spiking Test formulation and Standard to Human serum. The pink color is started developing immediately after addition of EDTA, TBA and HCL (Before keeping the sample at 80 Degree for 20 mins) The pink color formation will be at the end of reaction or initial stage-can anyone provide the reason.

2. We have observed pink colour based on the concentrations after 5.0 mins (Light-to dark pink) but we are unable to get linear absorbance at 532nm - can anyone provide the reason.

3. Currently anyone working on this project?
