I am planning to do Me-DIP sequencing for my project, however i am encountering some problems. So the NGS libary Prep kit that i bought (Active Motif) only allows dsDNA as input. However, after methylated-IP process the DNA are mostly ssDNA. I was wondering if anyone knows any kit that enables to conver the ssDNA to dsDNA for the librabry Prep. I have encounter a few kits that allows end repair+dA tailing + adapter ligation after DNA fragmentation but prior to the Methylated-IP conversion, however i am not sure how does these kit actually helps in converting the ssDNA to dsDNA. Any advice would be much appreciated.
