We are trying to use frozen, OCT-embedded tissue blocks to recover live enzymes. We specifically would like to look at tumor proteases from tissue blocks collected from patient samples. Recovering live cells from the blocks seems extremely unlikely, but we thought the proteases may still be active. However, none of us have worked with frozen, OCT-embedded tissue blocks before, so we are unsure if we are properly handling the material. Does anyone have advice on how to recover active proteases from frozen, OCT-embedded tissue blocks?
