We are trying to use frozen, OCT-embedded tissue blocks to recover live enzymes. We specifically would like to look at tumor proteases from tissue blocks collected from patient samples. Recovering live cells from the blocks seems extremely unlikely, but we thought the proteases may still be active. However, none of us have worked with frozen, OCT-embedded tissue blocks before, so we are unsure if we are properly handling the material. Does anyone have advice on how to recover active proteases from frozen, OCT-embedded tissue blocks?
how did you freeze the tissue blocks? How were they stored? How old are they?
Achim Recktenwald Sorry for the delay, I was waiting for information from the vendor (Origene) from which we we ordered the frozen, OCT-embedded tissue blocks.
1) I couldn't find out a specific protocol they used to freeze down the blocks, but they told me the blocks were placed in a labeled cryomold with OCT and frozen using vapor phase liquid nitrogen. The OCT-embedded sample was then removed from the mold and placed in a pre-cooled Cryosette.
2) It was then immediately placed on dry ice to be transported to and stored in a -80° C freezer.
3) The blocks are 10 years old.
