Has anyone experienced rapid decline in the dengue virus titre after harvest ? The titre drops to zero in 3 to 4 weeks after harvest.I work on clinical isolates that have been grown to P2 and onwards. I store in -80 after harvest. Kindly advise if there is any way to prevent this.
In this work, beta-carbolines harmol and 9N-methylharmine were found to have antiviral activity against DENV-2 virus in Vero cell cultures, causing a reduction in the number of infected cells and in the production of viral progeny.
Avoid the use of beta carbolines and methylharmine

Are you harvesting your supernatant with 20% FBS final volume for stability? What cell lines are you using for isolation?
Hi , the supernatant harvested is of 2% FBS in EMEM and I use vero cells.
