My protein is of molecular weight below 20 kDa. I transferred it for 30 min at 90V but could not see distinct bands i.e. smear type on Ponceau staining.15% gel was used and was run half of the total length of gel.
Once again. A very late answer. If you have not figured out your solution yet, yes your transfer should be longer. We typically transfer for 90V for two hours. If you are near the end of a work day, transfer for 30V overnight on ice. Make sure your transfer buffer contains SDS as well.
