I have no experience with qPCR. My supervisor asked me to order gBlock (gene block) as standard for qPCR. We are working with 4 different enzymes (Acid phosphatase, alkaline phosphatase, laccase and amylase). She told me to get the sequence for enzymes from an organism which is very common in nature. Someone in our lab worked for acid phosphatase enzyme and he used the sequence of Morginella sp. (a Bacterium). When I align the actual sequence and gBlock sequence, i found mismatch base pairs at few points. I thought that gBlock company might have done that, but they said they do not alter sequence. Now i am confused how to choose gene sequence for gBlock preparation. It will be great if you could help me.
