I have treated BALb/c mice with a therapeutic drug and would collect the spleens of these animals and check the APC population of Cd11c, Cd11b and Cd8 alpha and eventually the intracellular cytokines IL-10, IL-12 and the phsphorylated protiens ERK and MAPK, all by flow cytometry. What should be the best way to proceed for staining? Also how can one possibly stain both extracellular and phophoflow proteins in the same sample. All antibodies are from BD.
