Our lab has recently acquired a sonicator (from Qsonica) with a cup horn attachment and I have been designated as the person to set it up.
I have not been able to find much information from the product website or from online resources and wanted to ask if anyone out there uses this equipment. If so what settings do you generally use (i.e. time, amplitude, pulse)?
The sample volume that we are working with is generally 100 uL and the lysates are generated from tissue, specifically sciatic nerve and dorsal root ganglion.
I used to sonicate bacterial cells using sonicator (Sonics, USA) with 56%
amplitude, 10 s ON and 10 s OFF pulses for 7 min under ice.
2X 15sec sonication burst (with a small pulse on ice) for a 100ul bacterial lysate works fine
for bacterial it is better conditions we are currently working with 70%Amplitude and 0.25 Sec pulse on for 10Sec on and 20Sec Off. for mammalian cells it is better to use 45% Amplitude 4Sec on and 15Sec. both are under ice-bath only.
for bacterial we used 70% Amplitude at 0.25 Sec pulse on for 10Sec and off for20Sec in ice-bath.
Thank you for your answers however I was hoping to find out a protocol specific to eukaryotic cells and mammalian tissue. Currently I am using ~50% x 5sec on/off for 3-5 cycles.
Phillipe D. O'Brien Hi Phillipe, I am also starting with this kind of sonication. Could you please tell me if the above sonication conditions( ~50% x 5sec on/off for 3-5 cycles) worked or did you further optimize it.
