I am using Norgen RNA/Protein extraction kit to extract rna and protein from cortex. Following the protocol, everything went fine, however, after I adjusted the protein ph (adding molecular water and binding buffer), my protein precipitate. huge amount that block the column. I have tried collect protein from superfluid through centrifuge, however, there's barely anything in the fluid.
Anyone have any suggestion for me? really appreciate
I have used this kit and is very sensitive. Kindly adhere to the manufacturers instructions. As per the pH precipitation, I think this has to do with the lowering of the pH. This is because solubility of protein is dependent on the pH. At higher pH, protein becomes negatively charged while at lower pH, protein is negatively charged and also get precipitated easily. Hence, I advice that you adhere and readjust your pH carefully and you will get result
