Protein extraction protocol for trees (better if conifers)?

Hello,

I'm having trouble extracting and spectroscopically measuring whole protein content for conifers (pine and spruce). I'm using a protocol meant for barley. It works well with barley. Does anyone have any suggestions or protocols to share?
