Hi,

I am transiently transfecting my CHO cells using the Lipofectamine-2000 protocol. I will then be subjecting my cells to a stress stimulus (mediated through the protein expressed on my plasmid) and looking at pERK induction. I have read that for pERK serum starvation is recommended prior to stimulation (16-24 hrs). I am wanting to know if I can conduct my transient transfection in parallel with serum starvation. For example, I have my cells sitting overnight in optimem+lipofectamine complexes which would enable for transfection and serum starvation. My worry though is that my cells will not be able to express my protein of interest in serum starved conditions.

Thoughts?
