what concentration of the buffer should I used inside the dialysis tube with protein and what concentration of the buffer should I used outside for the dialysis purpose?
Can i use the same buffer concentration i.e 0.05M phosphate buffer with protein and outside the dialysis bag for dialysis?
Dialysis in protein purification is often used for buffer exchange (e.g. to make a sample compatible with a different column chemistry, remove imidazole, remove salt, etc.). In such a case the outside buffer will be whatever you want it to be, within reason. If the buffer concentration (e.g. Tris, HEPES, phosphate), is too low then it may take numerous exchanges and considerable time to alter the pH of the sample. Additionally, some proteins are unstable upon the removal of salt, so dialyzing the salt out of a sample may cause a protein to precipitate. I've heard it recommended to include ~150mM salt in the storage buffer to avoid such precipitation, but I have often found it unnecessary.
