Hello there

I am looking to use propidium iodide to make a yeast viability cell count using disposable haemocytomers and counting the cells manually under a microscope. The microscope in question is down a hallway from my lab, and is not inside of a flow hood or fume hood. I have many samples, so I will be counting for about 10 hours.

I am having some difficulty understanding the safety of handling dissolved propidium iodide outside of a fumehood. Most sources report that solid PI should always be handled under a fumehood, but accounts seem to vary with regards to the dissolved form. Most sources state that vapours should not be breathed, but with a boiling point of 220 degrees celcius, I assume that vapour formation is negligible.

If anyone has experience using PI in similar circumstances, I would appreciate any feedback! :-)
