Hi, I want to amplify the promoter region sequence in 3 species in the same Jenus?
what i have already done:
designing the primers according to the Ref. Seq. for 2kbp Upstream the CDS region.
amplified the promoter from species 1 and 2 using HiFi polymerase.
the sequences' identity was very high from 1 and 2 (95-99%).
But, the problem with species 3 no band in the length of 2000
I re-designed the primers until i get the required length band
But after sequencing the sequence identity is too low （40-50%）
what can i do to amplify the promoter sequence from species3?
