Im working on production cellulose from isolate of Actenobacter which is im sure it produce cellulose, also im using a optimum conditions temp, media and static incubation , but there is growth of bacteria without cellulose film for more than 20 days
could anyone advice please

Dear Ali;
My suggestion to you is first check your culture growth on agar media ( I used the Gluconacetobacter xylinus (ATCC® 700178™) strain )and collect the cultures on the agar first, and corn steep liquor (CSL)-glucose medium as growth medium . I advice you to sterilize the glucose, CLS and other separately before mixing your growth media. I collected my cellulose mats after 12 day at least 1 cm thickness. Reference is below.

I don't know what conditions you use to produce cellulose but I used Hestrin–Schramm (HS) medium with different carbon sources to produce cellulose by Gluconacetobacter in a 7-day fermentation at 30 °C with 150 rpm shaking.

As mentioned in the earlier responses, sterilization is very important. I had used coconut water as the growth medium for Bacterial cellulose. Initially, I was unable to obtain any cellulose when I had skipped sterilization of the medium. However, after sterilization, I was able to obtain a cellulose sheet of considerable thickness at room temperature. Hope this helps.
Good day! The general approach is:
1. Mix the medium
2. Sterilize medium and flasks
3. Inoculate fresh bacteria (up to 3 days)
4. Inoculate with loop, steriled by fire
Modern commercial bacteria strains are dedicated to the speed and effectivity of BC synthesis, so something will be growing even in the worth medium. So, maybe your bacteria isolate do not synthesize cellulose, or maybe some chemicals are inhibiting process.
Can i use peptone buffered water instead of peptone in bacterialcellulose production
Naheeda Gul It is possible, but remember about the 0.5% content of peptone in the final solution.
