I am trying to design primers for q-PCR but i have a doubt if i can use same gene of different organism eg : Can i use mouse TNF-alpha instead of human TNF-alpha.

Can you do qPCR on the same gene in multiple species? Yes. Sometimes you can even use the same primers, if the sequence identity is high enough.
Can you use the same gene as an internal reference in multiple species? Maybe not: frankly, you can't always use the same reference genes in a single species (what works in liver won't necessarily work in muscle, etc).
So in this instance you'd need to validate genes for each scenario independently.
But at present, you haven't really provided enough detail.
