Since both commercially synthesized primers and siRNA are oligos of 18-25 nt with different use; one for amplification and other for knockdown of gene. My query is that whether siRNA can be designed and synthesized similar to primers?
Commonly used primers are DNA primers, few companies synthesize RNA primers also. it is preferable to have an expression system in place so that you have control over the extent of knock down
if you still want to use synthesized oligonucleotides, RNA oligos can be used, other oligos that are used for silencing are Peptide Nucleic Acids (PNA) ; morpholinos; these have higher affinity to the RNA and unlike RNA they are resistant to degradation by nucleases.
