I have difficulties understanding how to make a solution diluted or concentrated.
For example: I have 1:1000 primary antibody and I want to make it 1:2000 , I dont know how to dilute it and which formula to use?
another example is : I want to make the solution concentrated , lets say I have 1:1000 primary and now I want it 1:500 concentrated.
I would really appreciate if you can help me out

1. Example: I have 1:1000 primary antibody and I want to make it 1:2000 , I don't know how to dilute it and which formula to use?
To make 1:2000 dilution from 1:1000 dilution, you may do it as follows.
2000/1000 = 2
which means you will need to do a 1:2 dilution, meaning 1 part of 1:1000 dilution (you may take specific volume of the solution (1:1000) which will suffice for your final dilution at the end) and 1 part of the diluting buffer to obtain a total of 2 parts. This will give you a solution (1:2000) which is diluted by half of the original solution (1:1000).
To summarize
So, if you need 10ul of 1:2000 dilution, you will take 5ul of 1:1000 dilution and 5ul of diluting buffer to obtain 10ul of 1:2000 dilution. Depending on your need (for the final volume of 1:2000 dilution) you could adjust the volume of the solution (1:1000) dilution and the diluting buffer.
2. Example: I want to make the solution concentrated , lets say I have 1:1000 primary and now I want it 1:500 concentrated.
A dilute solution is one in which there is a relatively small amount of solute dissolved in the solution. You cannot concentrate a dilute solution. To make 1:500 dilution, you cannot make it from 1:1000 dilution. You will have to use a solution of a higher concentration i.e., from the solution of a lower dilution say, 1:100 or 1:200 or you may make 1:500 dilution directly from the stock antibody vial.
