has anyone had to deal with cell lines that proliferate at 33 degrees and differentiate at 37-38 degrees (14 days needed) celcius?
The cell line im dealing with are SVI podocytes (CLS). i initially tried differentiation induction at 37 degrees but no differentiation was observed. i then tried temperature at 38.5 degrees but i see lots of cell death, and as at day 10 of differentiation, only a small portion of the cells look dfiferentiated.
i have heard that podocytes cell lines are extremely sensitive, for example: if any of the cells were to be overgrowned previously, these cells would lose its property and become resistant to differentiation.
any advise would be greatly welcomed!
