For virus isolation from raw fruit samples, the solution after precipitating with Polyethylene Glycol 8000 is becoming very thick and so it is not being feasible to use it for injection filtration using the 0.2 um filter, leading to sample loss. Can you suggest any alternative or better method to overcome this difficulty?
Dear Chayan,
in order to overcome several barriers due to virus isolation, we have developped an alternative method for capturing and concentrating viruses based on the Magnetofection technology.
It avoids laborious ultracentrifugation steps, or long and stressful on-column purification or laborious PEG precitpiation procedures.
The technology is based on Magnetic particles. In brief:
The procedure is really simple and do not need multiple steps ; simply add Mag4C reagents into the tube and place the tube on magnet. In this way, risks from bio-hazard are reduced as well as time to manipulate virus (viral particles stay intact).
Viral particles can be kept in complexion with magnetic nanoparticles and directly used for the Magnetofection procedure (enhancement of infection or transduction by magnetic field), or eluted and stored. Moreover, Magnetofection is totally compatible with in vivo so that captured virus can be directly injected.
