After an expression plasmid transformation into E coli, which event will come first?
Replication or expression?

after transformation we are choosing culture and mass culture so here without replication, impossible; and second thing u might have been experienced without adding promoter inducing factor for the gene of interest expression will not start. like IPTG after adding ly expression is starting and cell cycle is arresting.

Replication will be the primary thing that will happen in the cells as expression will only be possible when u induce some inducer as earlier mention like IPTG. So until you add some inducer, replication will be the only molecular event that will take place in the bacteria !!!
I agree with above answers. However gene of interest is under inducible promoter then you need to induce with inducer like IPTG. But in case of selection gene it will express simultaneously.

Replication is totally independent of expression of transgene and also its a natural property of any vector. In case of expression vector mostly the gene is under inducible promoter so it will not express until you add inducer like IPTG. But the other gene with like antibiotic selection gene which is naturaly present in vector is spontaneously expressed once it goes inside of the host cell. Or simply you can say that both function are present together for non - recombinant vector or cloning vector.
Replication and Expression are two different process, expression of gene is dependent on presence of regulatory sequence, without that it will not express. While, replication is for growth and development of plasmid, there is no relation with replication and expression of gene of interest. However, the level of expression may increase or decrease depending upon the increase/ decrease of cell number.
Replication first occurs so that the the copy number of plasmid increases and then expression occurs.ofcourse in this case you must add an inducer like IPTG in order to expression begins.
