Dear all
I have crude extract of several plant samples.the previous extract done by different solvent with different ratio(liquid/liquid).now I want to detect the content of alkaloids,phenols,flavonoids, tannine and steroid question is that:
1_Does each procedure require a certain solvent to dissolve the crude or it can be done simply by prepair solvents mixtuer similar to the mixtuer of solvent that had been used before in extraction process.
2- anybody have simple and fast protocol to do all tests above?

You can screen for phytochemicals using the same crude extract you have obtained. NB. Some method may require you to dissolve it in different solvent.

For quantitative analysis, the whole sample not the extract will be in use.

As per seen in your methods for detection test for flavonoids,tannine,saponineand others you were using only water as solvent .Question is can I use my previous extract solvents mixture or is must use water only
Yes, we used water and in the case of terpenoid, chloroform was used because each phytochemical is solvent dependent. You can as well use solvent ratio once u are able to tune the polarity...but honestly, I will suggest that you follow already established procedure as reported in the attached publication.

There are several standard protocols for cualitative analysis during the initial stages of phytochemical studies. You can follow only one or the combination of several methodologies
