Hi..I would like to ask some questions to all the researchers who had experience using L-NAME ( L-NG-Nitro arginine methyl ester ) to induce hypertension in rats model. I had given L-NAME to my animal for 8-10days via oral gavage every day in the morning and then took the BP shortly after oral gavage. I found that, the drug was unable to cause HPT in my animal..So, currently i want to troubleshoot whatever i could..I afraid that the drug take some time to give action..but somehow, i didn't found the answer...Please enlighten me..Thanks in advance
This is actually strange. L-NAME is reported to have acute and chronic effects. So you should be able to see effects on blood pressure shortly even after the first administration. However the L-NAME dose is important. A dose too low may increase NO production for a feed back positive loop. Please find two papers which may be of help (both are free full texts). My suggestion is to try a single dose-response and then use the dose inducing BP changes for the repeated administration.
Thanks for your response..FYI, i gave 60mg/kg/day via oral gavage to my animal about 8-10days..is it the dosage considered low? and is it the mode of administeration also influenced the effect of the drug?BTW, I actually want to induce HPT in pregnant rats..
L-NMMA can quickly disappear from the plasma but has a prolonged pharmacological effect.
