I want to perform an IVT by plasmid. It is necessary to use SC form than OC form or I can use both forms for linearization brfore IVT?

it has been many years since I did IVT from plasmid but you usually have to linearise and purify to do IVT
That is because non linear plasmids have secondary structure that can prevent efficient IVT
