ELISA seems like an easier method than Western BLot, but most papers that involve detection of p38 phosphorylation or P38 MAPK activity involve the Western Blot technique. Does anyone know of why this might be? Does anyone have recommendations for which method is better (ELISA vs Western BLot)?

The choice of detection approach in part depends upon the source and availability of the sample and the quality of the antibody available. If the source of sample is not limiting and the antibody is well-characterized and authenticated, all three approaches (which are complementary) could result in similar outcome. However, if the source is limiting (e.g., clinical sample), but the antibody is authenticated, the preferred method could be either ELISA or immuno-fluorescence. When the sample is a cell line-derived and antibody is not authenticated, Western blotting is preferred choice.
Thank you for your response Divaker Choubey. I'm working with MEFs/cell culture. Does this mean Western blot is preferred? Could i still have a similar outcome with ELISA?
Divaker Choubey I forgot to mention RT-PCR is also used as a method.
