I want to do RNA seq and I am starting from small number of sorted cells. I though using the SMARTer Stranded Total RNA-Seq Kit - Pico Input Mammalian in combination with NucleoSpin® RNA XS seems a good way to go. Unfortunately, I can find very little information on NucleoSpin® RNA XS - and I was wondering whether anyone has used them.
This is a Macherey-Nagel kit corresponding to the Qiagen RNAeasy micro kit, you can elute your purified RNA in a very small volume (10 µl if I remember correctly). Like all types of column preparation, you'll loose all the small RNAs (miR and their precursors), but if it is not your concern, then I think you can use this kit without any problem. Dry the columns very well after the ethanol wash (add an additional 2' >10000 rpm centrifuge with no buffer after the last ethanol wash) to be sure to avoid ethanol carry-over, and try to be very careful to avoid guanidine thyocyanate transfer in the first steps. I have used this kit to extract my RNAs from small amount of tissue and I have done a RNAseq experiment after. It was perfect, just follow the protocol but remove the carrier RNA step !
