I am facing one problem in case of quantification of nucleic acid conc. wit udrop plate which pathlength is 0.5 mm. Whether it is necessary for me to quantify the concentration in respect to 10 mm pathlength.
The concentration is the same whether you measure it at 0.5 mm or at 10 mm pathlength. What changes is the actual OD. If the instrument gives you directly a concentration reading (based on a preset extinction coefficient for 0.5 mm or whatnot) you do not need to convert anything.
