Dear fellow researchers,

I want to do pull down experiments of RNA associated with the protein of my interest in human ES cells. It shuttles between the nucleus and the cytoplasm. The protocol that I have works for other cell types like HeLa and HEK, it does not seem to be working for ES Cells. I am guessing that's because the ES cells have a much higher nucleus/cytoplasmic ratio and they way I end up handling them, just isn't working for me.

Any tips/suggestions are welcome.
