I am using Trizol LS reagent for RNA and DNA extraction from Grape-vine leaf tissue, but I couldn't find any RNA and DNA. I found big pellet and a bit it is hard to dissolve. I check the quality using nono drop and it seem there are some RNA but poor quality. When I check it using bio-analyzer, there is no RNA at all. Can any one help please?
hello Kiflu, try to be less greedy while taking aqueous phase . it is not necessary that you get a big pellet. quality is more important . even in small pellet you will get good amount of RNA.

extraction of DNA and RNA there are different ways for that sometimes genomic dna extracted by sucrose buffer and it is a difficult and results not satisfied but I preferred that extracted by specific kits from these companies specific in this field.
