I am running a sample through our HPLC which the peaks are unknown due to it being a new molecule. However, I ran the sample today and the entire spectrum was in the negatives. I have seen negative peaks before but never the entirety of the spectrum negative. This hasn't happened since this run, but I have observed more negative peaks than before. I attached a photo below.
I am running a Waters system with a C8 column, 40 uL injection, and 0.8 mL/min flow rate. My methods are as follows: 0 to 15 minutes gradient 60% to 90% MeOH in Water then 15 to 20 minutes isocratic 90% MeOH in Water.
The signal noise is very small. Probly the system used autozero function during elution of some impurities.

Can you try to increase the sample concentration, if the concentration is very low it could be covered in the noise. otherwise you may be having too much noise with your system

*Change detection wavelength
*Dissolve your sample in mobile phase

Thank you all! I think my sample concentration was just too low. I increased the concentration and the issue seemed to be solved.
However, I am relatively new at using the HPLC, are there any tips for lowering background noise?
