Measure the total levels of gangliosides in serum. For melanoma tissues, you can do confocal for looking into various types of gangliosides. Gangliosides are immunosuppressive. I have worked on human melanoma and you want to search for its application in mouse system. Refer to work done by Dr. Ravindranath and Dr. Finke.

Debarshi, do you also want to distinguish melanoma from melanocytes, or is a melanocyte-specific marker ok (like MITF, tyrosinase, TYRP1)? All of these are ok (in tissues, not sure about serum) for a well-differentiated melanoma - which they may be, even in white mice (your Balb/c's are white, right?). Albino melanocytes still make all the same proteins. If some melanomas dedifferentiate in your model then you'll need a marker that is also in melanoblasts, which might be DCT or PAX3. MAybe S100 too. Vince Hearing at the NIH (USA) used to have good anti-mouse antibodies to nearly all of these but he retired recently, so I don't know what happened to his antibodies. The silver or PMEL melanosomal matrix protein is the target of the NKI/beteb and HMB45 antibodies, also gp100, but they are raised against human, so not sure how good in mice. Also will be less positive upon dedifferentiation.
Mam, I'm using White Mice (Balb/c) 5-6 weeks old, actually I'm bit confused with the specificity of the markers like if i use S100B protein as a marker in my experiment so whether this marker will be specific for mice model or i can say that; will it show the same specificity as it has shown in humans?
S-100 is not a reliable marker for Melanoma at all! It is old fashioned, not specific, and works different depending on its clone...Believe me, as a pathologist with 17 years experience))
s100 sensitive but not specific hmb45 specific but not very sensitive , melanin A another name MART1 a bit more sensitive than hmb45. none are specific for melanoma as benign nevi are alo staining same way. i am not aware of any working serum markers for melanoma.
Debarshi, you asked if these markers will also be specific in mice.
All of the melanosomal proteins will be specific for melanoma (plus naevi and melanocytes), as opposed to tumours of other lineages, in any animal. These include tyrosinase, TYRP1, MART1/MELANA, DCT and PMEL/PMEL17, (the antigen for HMB45 etc in humans). MITF would also be very specific - this is a melanocytic transcription factor, and is abundant enough in cells to see it. The S100 family are also in glial cells and their tumours, so not quite so specific, even with a good antibody. So you can check the companies for anti-mouse antibodies against any of these.
