I would like to check the bio-compatibility of a polymeric material. what are the biochemical/toxicological test or any other method that I can perform in order to prove the bio-compatibility.
Any suggestions on cell line culture or any other method will be highly appreciated.

Dear Sir. Concerning your issue about how to check the biocompatibility of a polymeric material. An in vivo method is described for screening polymeric materials for biocompatibility. The test is based on grading acute and subacute tissue reactions at 7 and 28 days, respectively, following implantation in rats. The method is reproducible and reliable. It is designed to provide uniform test criteria for biocompatibility assessment in the early phases of the development of surgical implant materials. Thanks Dr. Isam for your insightful answer with nice recommended articles. I would like to know your view or insights on the question that I would like work with only in-vitro experiments not in-vivo at the moment as working with Animals always includes whole lot of ethical issues.

First of all, You have to do viability test using L929 cell line (fibroblasts). Use Trypan Blue or other staining. You can use MTT test, but You have to be carefull if use cyclic compounds, they give false-positive results. The next step is to use any other cell line which is closer to tissues which are intended to contact with this material. If this material will show good biocompatibility, apply primary cells (if possible), and the final step is the tests in vivo where You can observe fibrousus or other capsules or their absent.

biocompatible polymers must have functional groups such as hydrophilic or lipophilic,commonly contain chemical
linkages such as anhydride, ester, or amide bonds
.These polymers degrade in vivo either enzymatically or
non-enzymatically to biocompatible and non-toxic
byproducts. It can be metabolized or excreted via
normal physiological pathways.

A 1.00mg of polymer can kept in a cylinder containing 100 ml of buffer solution in different pH values at 37 0C without stirring. A released sample periodically reserved and analyzed by UV. Spectroscopy at specific λmax used to determine the amount of the hydrolysis unite. A calibration curve must construct with a software built in the computerized UV. Spectrophotometer, the amount of the hydrolysis amount can determined directly from the software for many measurements with time,
