Does anyone have the similar issue? A line across the denature SDS gel shows up after staining with Coomassie blue and there are many lines in every lane if you take a closer look. This gel had 15 wells, I only used four lanes in the middle of the gel, so the wells on the side should not contain any proteins. This issue has happened to my labmates several times as well. We used fresh buffer every time we ran a gel. We are not overloading samples. Is there something wrong with the Coomassie blue staining solution or loading dye? or is it the gel solution? Thank you!
My first instinct was to say your gel is expired. However, I came across this thread from 2018 where a similar problem was described. The consensus seemed to be related to pH shift during the run (due to high temps). I imagine this would be resolved by slowing things down a bit, or running the gel in cooler conditions.
