I started working on the analysis of multivitamins in Gummies by HPLC using DAD. Unfortunately, I don't find enough supporting material about the project.
If someone has already worked on the same project or isfamiliar withit, mayIknow what is the best diluent to dissolve Gummies at room temperature for sample preparation? What are the ideal chromatographic conditions including mobile phase, column, injection volume, etc?

Use water and methanol as diluent, and use phosphate buffer and acetonitrile as mobile phase, column C8, detector UV visible. Always use water soluble things in such diluents which have more parts are aqueous like water or phosphate buffer and for organic use diluent which have more parts are organic like methanol acetonitrile as a solvents.
The real question is 'what vitamins' and 'what concentration'. Because you may have both fat soluble and water soluble vitamins you may have to have 2 separate extraction procedures. In addition you will have to ensure that your formulation (dummies including dyes and flavors) do not interefere.
@Saeed Ayaz Khan: I have tried the poster published by "South Dakota State University" but it didn't work. I have written to the author and waiting for a response. The Gummies at hand didn't dissolve in MeOH:Water (1:1) at room temperature despite extended hours of shaking.
@Bruce: I am trying to determine both water soluble and fat soluble vitamins and the specification about their concentration is not provided. It's the reason that I am trying calibration curve to get the concentration in unknown.

You have to know the composition of the gummies. There are 3 basic types of gummies - there are gelatin gummies, which will dissolve (slowly) at room temperature. There are pectin gummies, which will never dissolve at room temperature and there are carragenan gummies which also will not dissolve at room temperature. I analyze gummies for terpenes and cannabinoids. We chop them into small pieces, put them in isopropanol/water and heat them to around 60 C with shaking, and even then the pectin gummies take a long time to dissolve. I do not know how hot you can go with the vitamins. I know that some of the vitamins are not very temperature stable but they must be somewhat stable in order to have been made into a gummy.
Also, as Bruce points out, you will have to extract the fat soluble vitamins into a non-polar solvent and water soluble vitamins into a polar solvent. We use the isopropanol because we get the added advantage of dropping out the sugars that are present; you will get this with methanol as well.
You almost certainly will have to use the method of known additions for the analysis, since the matrix definitely causes problems using HPLC.
