Does anyone have experience with mouse leukocyte transendothelial migration assays? I am trying to set up such a system, but am short of an appropriate cell line. Could anyone suggest a cell line or method of isolating primary endothelial cells? How do the different endothelial cells differ in terms of their permeability and ICAM-1 expression? Thank you very much!

The Stamper-Woodruff assay is something you might want to consider (e.g. http://www.sciencedirect.com/science/article/pii/S0022175906000755).
In terms of cell lines the gold standard here is usually HUVECs and can be purchased from many vendors (in the past I have used Lonza: http://www.lonza.com/products-services/bio-research/primary-and-stem-cells/human-cells-and-media/endothelial-cells-and-media/huvec-human-umbilical-vein-endothelial-cells.aspx)
