Hi, currently I am working to isolate DNA for 16s RNA sequencing (metagenomic purpose) from Beaver guts. I isolate with SDS-based DNA extraction method (Zhou et al. 1996. DNA Recovery from Soils of Diverse Composition). I obtain the DNA, but it's solution color were brownish. So I try to remove all impurities in DNA solution using Mo-Bio PowerClean Pro DNA Clean-up kit. Sadly, not only the impurities were gone, but the DNA as well. The DNA final concentration measured using Nanodrop is around 5 to 8 ng/ul (it was more than 100 ng/ul but it could be biased because of impurities). Anyone have similar relust before with this PowerClean kit? Can you suggest method to clean up the DNA?
