I suggest the widest possible collection of genotypes and growth analysis during ealry development. doses are tested with conducimetry and vary around 10 dS/m http://www.sciencedirect.com/science/article/pii/S025462990600161X The equivalence to molar units is approximately I (mol/L) = 1.6 10-2 × EC (dS/m). Seawater ranges above 50 dS/m. Plants sense conductivity more than molarity

1. You should conduct the preliminary expt.. on wheat to find out which dose is proper.
5. For wheat cultivation 15-22 degree temp. is better for proper growth and development. Under high temp. growth is inhibited.
you should contact the published work on salinity tolerance of wheat,to select the proper salinity levels. you could start at ,20,40 ,60mM NaCl. this could be easily done using molar concentration of NaCl . remember 10mM NaCl=1ds-m. chlorophyll could be determined using the method of Goodwin(1965). proline could also be dtermined using the method of BATES ET AL,1973.
1. Use 10, 20, 50 100 and 200 mM concentrations.
2. For estimation of chlorophylls follow Arnons Method. Extraction in 80% acetone, reading absorbance at 663 and 645 nm.
3. Estimate Proline following Bates et al. (1973).

salt stress affects leaf growth, so you could look into leaf histology measuring cell size parameters. The following article is a possible suggestion:

Leaf firing is another easily measurable visual trait to screen for the effects of salt stress.
It will be better if you first check the response of a diverse set of genotypes in petri-plates then go for hydroponic culture.
In case of histology analysis it will be better if you go with roots.
Well, there could be few other stress indicators which may help to evaluate the plants response to salinity stress too; e.g. chlorophyllus and carotenoids, electrolyte leakage, lipid peroxidation
You can also measure antioxidant enzymes like SOD, CAT, GPX, APX and MDA. Then you find the ratio of antioxidants to MDA among your genotypes.Then you find the correlation between seed yield and your finding ratio with statistical methods.It will show you resistant genotypes.
