Hi all,

I recently got a gene cloned into pENTR223.1.

when i sequenced it and compared to the annotation, i saw that the attL1 site, instead of ending with GTTG, ends with a G (missing either GTT or TTG, depends how you look at it...)

performing LR reaction on the computer didn't work with these 3 missing basepairs (while working with the complete attL1 site), and the in-vitro reaction did not work as well

does anyone know if indeed the reason is these missing 3 bp?
