You can use any minimal medium, but the only source of Carbon must be ACC, so I don´t recommend to use glucose, good luck

You might use ACC as the only Nitrogen source too. Quantify ACC-deaminase products after the assay to make sure you are not having interference from biological nitrogen fixation.

I used a minimal medium to test ACC-deaminase producing ability of a PGPB. If you still need to have a minimal medium composition, could you reply me your email so that I can send it to you?

ACC deaminase activity on the sterile minimal DF (Dworkin and Foster) salts media (DF salts per liter: 4.0 g KH2PO4, 6.0 g Na2HPO4, 0.2 g MgSO4.7H2O, 2.0 g glucose, 2.0 g gluconic acid and 2.0 g citric acid with trace elements: 1 mg FeSO4.7H2O, 10 mg H3BO3, 11.19 mg MnSO4.H2O, 124.6 mg ZnSO4.7H2O, 78.22 mg CuSO4.5H2O, 10 mg MoO3, pH 7.2) amended with 3 mM ACC instead of (NH4)2SO4 as sole nitrogen source (Dworkin and Foster, 1958; Penrose and Glick, 2003). However, I'm very surprised to see that it doesn't contain any source of Calcium. Is it normal ? Could it impede the growth of bacteria (PGPB) ?
