Hi everyone. Does anyone here ever did a study on benthic microalgae and quantify the microalgae using coverslip method? What calculation did you used to find the cell abundance/density per m2 @cm3 of your samples? Any references to that?

In my case, I collected the mud samples using syringe and extract top 1cm of the mud for diatom identification and quantification. I quantify using light microscope under 40x magnifications.

I've been using formulas from several references to calculate the cell abundance per cm3. But I want to ask if any of you has been using the same method as I am. Calculating the cell using coverslip instead of sedgewick rafter and other method. So I can have a better idea if my calculation is accepted.
