What method of transfection do you generally use in your lab? We often transfect cells, unfortunately we have not transfected H295R cells before but have transfected other whole cell models such as LNCaP cells. We use transfection reagent and transfect according to the manufacturer's instructions. Depending on the toxicity of the reagent you are using it may cause the cells to lift. As H295R cells can be slightly picky, if using a reagent it may be beneficial to plate into CellBind plates to ensure adherence throughout the experiment. We use XtremeGene Transfection Reagent and include treated, untransfected cells as a control to ensure the transfection has taken place. This would allow you to compare results of with and without the enzyme which was introduced.
I see that people have use Lipofectemene which is extreamly expensive. I was wondering if anyone had any luck with a cheaper reagent. Normally we transfected cells with Calcium phosphate but i dont think that would work for that.
we have not yet collected data for plasmid transfection, but our Viromer BLUE works well for transfecting siRNA in these cells... I would suggest to try Viromer RED or Viromer PLASMID as it's the same technology.
